DESCRIPTION

SH3BGRL2 belongs to the thioredoxin (TXN; 187700) superfamily, but it lacks the CxxC motif essential for redox catalytic activity (Mazzocco et al., 2002).

CLONING

By searching an EST database for sequences similar to SH3BGR (602230) and SH3BGRL (300190), followed by RT-PCR of HeLa cell total RNA, Mazzocco et al. (2002) cloned SH3BGRL2. The 3-prime UTR of the transcript contains multiple AU-rich degradation signals and 2 polyadenylation sites. The deduced 107-amino acid protein has a calculated molecular mass of 12.3 kD. It shares 63% amino acid identity with the N-terminal region of SH3BGR and 63% and 39% identity with the shorter SH3BGRL and SH3BGRL3 (615679) proteins, respectively. SH3BGRL2 has a C-terminal proline-rich sequence with SH3 domain- and HOMER (see 604798) EVH1 domain-binding motifs, and it is characterized by the presence of a thioredoxin-like fold. However, SH3BGRL2 lacks the cysteines necessary for catalytic activity. Northern blot analysis detected variable expression of SH3BGRL2 transcript of about 5 and 3.5 kb in all human tissues examined, with highest expression in brain, placenta, liver, and kidney. Fluorescence-tagged SH3BGRL2 was expressed in the nucleus and perinuclear region of transfected COS-7 cells.

Mazzocco et al. (2002) determined that the SH3BGRL2 gene contains 4 exons and spans 72 kb.

MAPPING

By genomic sequence analysis, Mazzocco et al. (2002) mapped the SH3BGRL2 gene to chromosome 6q13-q15. Gross (2014) mapped the SH3BGRL2 gene to chromosome 6q14.1 based on an alignment of the SH3BGRL2 sequence (GenBank GENBANK AF340151) and the genomic sequence (GRCh37). ... More on the omim web site
