Phosphopantothenate--cysteine ligase (PPCS)

The protein contains 311 amino acids for an estimated molecular weight of 34005 Da.

 

Catalyzes the second step in the biosynthesis of coenzyme A from vitamin B5, where cysteine is conjugated to 4'-phosphopantothenate to form 4-phosphopantothenoylcysteine (PubMed:11923312, PubMed:12906824, PubMed:29754768). Has a preference for ATP over CTP as a cosubstrate (PubMed:11923312). (updated: Sept. 18, 2019)

Phosphopantothenoylcysteine synthetase; ppcs

DESCRIPTION

Biosynthesis of coenzyme A (CoA) from pantothenic acid (vitamin B5) is an essential universal pathway in prokaryotes and eukaryotes. PPCS (EC 6.3.2.5), one of the last enzymes in this pathway, converts phosphopantothenate to phosphopantothenoylcysteine (Daugherty et al., 2002).

CLONING

By searching for sequences similar to PPCS from Streptococcus pneumoniae, followed by PCR of a brain cDNA library, Daugherty et al. (2002) cloned human PPCS.

GENE FUNCTION

Daugherty et al. (2002) showed that recombinant PPCS functioned within the CoA synthetic pathway. PPCS used ATP for the activation of substrate in the ligation reaction 4 times more efficiently than CTP. Daugherty et al. (2002) verified the function of PPCS by complementation in E. coli. Incubation of PPCS, PPCDC (609854), and the bifunctional enzyme COASY (609855) with the necessary substrates and cofactors reconstituted the 4-step biochemical transformation of phosphopantothenate to CoA.
